BDNF基因的MSC自体移植对视网膜损伤的修复作用

  【目的】 探讨转BDNF基因的间充质干细胞(MSC)自体移植后对视网膜激光损伤的修复作用。【方法】 激光损伤视网膜的32只青紫兰兔随机分成两组。一组左眼注射生理盐水,右眼注射MSC;另一组左眼注射MSC,右眼注射转BDNF基因的MSC。术前、术后分别检测兔的眼压、B超、ERG、眼底照相,及视网膜病理检查。【结果】 术前、术后眼压和B超检查未见明显变化。ERG、眼底照相和视网膜病理检查,MSC自体移植组与生理盐水组对比视网膜损伤轻;转BDNF基因的MSC自体移植组比单纯MSC移植组视网膜损伤更轻。【结论】转BDNF基因的MSC和单纯MSC自体移植后对视网膜损伤均有修复作用,且转基因的MSC的修复作用更强。

  Mesenchymal Stem Cells Transfected with BDNF Gene

  for Autologous Transplantation to Repair of the Retina Damage

  [Objective] To eveluate the repairing function of mesenchymal stem cells transfected with BDNF gene for autologous transplantation to the retina damage. [Method] 32 rabits with laser damaged retina were divided into two groups randomly. The left eyes in group one were injected saline, and the right eyes were injected MSCs. The other group, the left eyes were injected MSCs, and the right eyes were injected hBDNF gene transfected MSCs. Pre- and post-operations respectively, IOP, B- ultrasonic, fundus photography, multi-ERG and retina pathology were examed. [Results] There were no significent changes in IOP, B-scan. But for ERG, fundus potography and retina pathology, the repair of damage retinas were better in MSCs group than saline group, and hBDNF gene transfected MSCs group was the best. [Conclusion] MSCs and BDNF gene transfected MSCs have repairing function to the laser damaged retina. And transfected MSCs’ repairing function was stronger.
